Past Project 2016-17: Effect of 2,4-thiazolidinedione on goat response to mastitis and milk fat - Study two

The objectives of the study were to assess in well-fed lactating goats the effect of 2,4-thiazolidinedione, a putative PPARgamma agonist, on: 1) milk fat production 2) the response to induced sub-clinical mastitis

Main findings.

Objective 1 - We determined that the daily injection of 8 mg/kg body weight of 2,4-thiazolidinedione in well-fed Saanen dairy goats dramatically reduced the NEFA and the BHBA in blood and increased glucose with consequent increase in adipocytes size. It affected also the expression of insulin- and glucose metabolism-related genes in the adipose tissue but had minor effect on mammary cells.

Objective 2 - The injection of 2,4-thiazolidinedione slightly improved the liver situation but did not affect the inflammatory response of mammary epithelial cells or macrophages isolated from milk. Data also indicated that 2,4-thiazolidinedione is not a PPARgamma agonist. 

Overall well-fed goats had a lower response to 2,4-thiazolidinedione compared to the same goats in less than optimal nutritional status.
